Comparing Different Supplements for Cell Culture, Expansion, and Cryopreservation

The researchers identified factors including immunomodulatory molecules (IL-10, IL-6, TGF-alpha, GM-CSF, G-CSF), the RANTES chemokine, and growth factors (IGF-1, VEGF-A, PDGF-AA, PDGF-AB, basic FGF, EGF, and TGFβ) that play roles in cell stimulation, differentiation, maintenance, and growth. These factors were found to be significantly higher in platelet-derived preparations compared to FBS.

The concentrations of soluble factors representative of the platelet present in HPL and PLS were higher than those in commercial FBS. HPL and PLS are supplemented at 5% in culture media, half the concentration of FBS at 10%. Due to the varying amounts of growth factors and biochemical components in these platelet derivatives, indicating that the choice of supplement can greatly impact cellular growth and development conditions. Also, pooling platelet lysate from hundred donors reduced batch variability.

The research also shows that using HPL animal serum-free media results in decreased operating costs and greater safety for the use of cells in advanced therapies.
