Blood derivatives have emerged as FBS alternatives, with human plasma, serum, and platelet concentrates (lysates or serum-converted) being studied for MSC-based cell therapy production.Platelet concentrates have become essential in MSC production, offering cost-effective, large-scale, and standardized supplement production. Human platelet lysate (hPL) is a powerful xeno-free supplement that promotes cell growth, survival, proliferation, and maintains the MSC phenotype.
In this study, the researchers thoroughly characterized hPL batches, focusing on growth factor content and stability, and compared them with human serum (hS) and FBS supplements. They hypothesized that hPL delivers essential factors that are absorbed by WJ-MSCs, enhancing their regenerative potential through the continuous secretion of pro-survival and immunomodulatory signals.
The researchers assessed hPL stability by examining the variation in soluble factor levels across different batches. They evaluated the growth kinetics and viability of WJ-MSCs cultured in media supplemented with hPL, hS, and FBS using a luminescence-based Cell Viability Assay. Additionally, they performed immunophenotypic characterization and assessed differentiation potential in cells expanded with these supplements. The study also involved monitoring the accumulation of soluble factors in the supernatants from WJ-MSCs cultured in the presence of hPL, hS, and FBS over a five-day period.
hPL is a suitable source of factors supporting viability, stability and potency of WJ-MSC and therefore constitutes an essential raw material that in combination with WJ-MSC introduces a great opportunity for the generation of potent regenerative medicine products.
